Chromosome disorders can be divided into two categories: abnormalities in chromosome number and chromosomal structural rearrangements. Because even small segments of chromosomes span many genes, chromosomal disorders are characteristically dramatic and often fatal.\r\n<h2>Identification of Chromosomes<\/h2>\r\nHow does one find out about their genetic make-up?\u00a0 What diseases, if any, do I have?\u00a0 What sort of chromosomes do I pass on to my progeny?\u00a0 One way we can find answers to these and other genetic questions is to have a karyotype. \u00a0 A karyotype is the number and appearance of chromosomes, including their length, banding pattern, and centromere position. To obtain and view an individual\u2019s karyotype, cytologists take a sample and allow the cells to grow for staining.\u00a0 Proper arrangement is done so a photograph the chromosomes and then cut and paste each chromosome into a chart, or karyogram, also known as an ideogram (Figure\u00a01).\u00a0 More details in karyotype preparation follows below in Career Connection.\r\n\r\n[caption id=\"attachment_1466\" align=\"aligncenter\" width=\"800\"]<img class=\"size-full wp-image-1466\" src=\"https:\/\/s3-us-west-2.amazonaws.com\/courses-images\/wp-content\/uploads\/sites\/198\/2016\/11\/28190018\/Figure_13_03_01.jpg\" alt=\"This is a karyotype of a human female. (credit: Andreas Blozer et al)[\/caption]\r\n\r\nIn a given species, chromosomes can be identified by their number, size, centromere position, and banding pattern. In a human karyotype, autosomes(non-sex) are generally organized in approximate order of size from largest (chromosome 1) to smallest (chromosome 22). The X and Y chromosomes are not autosomes.\u00a0 During the naming of Down syndrome to trisomy 21, it was discovered that chromosome 21 is actually shorter than chromosome 22.\u00a0 Not wanting to change the name of this important disease, chromosome 21 retained its numbering, despite being the shortest set of chromosomes. The chromosome \u201carms\u201d projecting from either end of the centromere may be designated as short or long, depending on their relative lengths. The short arm is abbreviated <em>p <\/em>(for \u201cpetite\u201d), whereas the long arm is abbreviated <em>q <\/em>(because it follows \u201cp\u201d alphabetically). Each arm is further subdivided and denoted by a number. Using this naming system, locations on chromosomes can be described consistently in the scientific literature.\r\n<div class=\"textbox key-takeaways\">\r\n<h3>Career Connection<\/h3>\r\n<h4>Geneticists Use Karyograms to Identify Chromosomal Aberrations<\/h4>\r\nAlthough Mendel is referred to as the \u201cfather of modern genetics,\u201d he performed his experiments with none of the tools that the geneticists of today routinely employ. One such powerful cytological technique is karyotyping, a method in which traits characterized by chromosomal abnormalities can be identified from a single cell. To observe an individual\u2019s karyotype, a person\u2019s cells (like white blood cells) are first collected from a blood sample or other tissue. In the laboratory, the isolated cells are stimulated to begin actively dividing. A chemical\u00a0 is then applied to cells to arrest condensed chromosomes in metaphase. Cells are then made to swell using a hypotonic solution so the chromosomes spread apart. Finally, the sample is preserved in a fixative and applied to a slide.\r\n\r\nThe geneticist then stains chromosomes with one of several dyes to better visualize the distinct and reproducible banding patterns of each chromosome pair. Following staining, the chromosomes are viewed using bright-field microscopy. A common stain choice is the Giemsa stain. Giemsa staining results in approximately 400\u2013800 bands (of tightly coiled DNA and condensed proteins) arranged along all of the 23 chromosome pairs.\u00a0 An experienced geneticist can identify each band. In addition to the banding patterns, chromosomes are further identified on the basis of size and centromere location. To obtain the classic depiction of the karyotype in which homologous pairs of chromosomes are aligned in numerical order from longest to shortest, the geneticist obtains a digital image, identifies each chromosome, and manually arranges the chromosomes into this pattern (Figure 1).\r\n\r\nAt its most basic, the karyogram may reveal genetic abnormalities in which an individual has too many or too few chromosomes per cell. Examples of this are Down Syndrome, which is identified by a third copy of chromosome 21, and Turner Syndrome, which is characterized by the presence of only one X chromosome in women instead of the normal two. Geneticists can also identify large deletions or insertions of DNA. For instance, Jacobsen Syndrome, involving distinctive facial features as well as heart and bleeding defects, is identified by a deletion on chromosome 11.\u00a0 The karyotype can pinpoint translocations, which occur when a segment of genetic material breaks from one chromosome and reattaches to another chromosome or to a different part of the same chromosome. Translocations are implicated in certain cancers, including chronic myelogenous leukemia.\r\n\r\nDuring Mendel\u2019s lifetime, inheritance was an abstract concept that could only be inferred by performing crosses and observing the traits expressed by offspring. \u00a0 Today\u2019s geneticists can actually visualize the chromosomal composition of an individual to confirm or predict genetic abnormalities in offspring, even before birth.\r\n\r\n<\/div>\r\n<h2>Disorders in Chromosome Number<\/h2>\r\nAbnormalities in chromosome number are the most obviously identifiable form of a karotype.\u00a0 Most of these are due to nondisjunction.\u00a0 <span style=\"text-decoration: underline\">Nondisjunction<\/span> occurs when homologous chromosomes pairs or sister chromatids fail to separate during meiosis. Causes that attribute to nondisjunction are misaligned or incomplete synapsis or a dysfunction of the spindle apparatus, facilitating chromosome migration. Nondisjunction rates increase with parental age.\r\n\r\nNondisjunction can occur during either meiosis I or II, with differing results (Figure\u00a02). If homologous chromosomes fail to separate during meiosis I, the result is two gametes lacking a that particular chromosome and two gametes with two copies of the chromosome. The incidence of having a fetus with trisomy 21 increases dramatically with maternal age.[\/caption]\r\n\r\nAn individual with the appropriate number of chromosomes for their species is called <span style=\"text-decoration: underline\">euploid.<\/span> \u00a0 In humans, euploidy corresponds to 22 pairs of autosomes and one pair of sex chromosomes. An individual with an error in chromosome number is described as<span style=\"text-decoration: underline\"> aneuploid<\/span>, a term that includes monosomy (loss of one chromosome) or trisomy (gain of an extraneous chromosome). Monosomy zygotes, missing any one copy of an autosome, fail to develop to birth because they lack essential genes.\u00a0 Most autosomal trisomies fail to develop to birth; however, duplications of some of the smaller chromosomes (13, 15, 18, 21, or 22) can result in offspring that can survive.\u00a0 Trisomic individuals suffer from\u00a0 an excess in gene dose. Individuals with an extra chromosome may synthesize an abundance of the gene products encoded by that chromosome. This extra dose of specific genes can lead to a number of functional challenges, often precludes development. The most common trisomy among viable births is that of chromosome 21, or Down Syndrome. Individuals with this inherited disorder are characterized by short stature, stunted digits, facial distinctions including a broad skull and large tongue, and significant developmental delays. The incidence of Down syndrome is correlated with the age of the mother.\u00a0 Older women are more likely to become pregnant with fetuses carrying the trisomy 21 genotype (Figure\u00a03).\r\n<h2>Polyploidy<\/h2>\r\n[caption id=\"attachment_1470\" align=\"alignright\" width=\"349\"]<img class=\" wp-image-1470\" src=\"https:\/\/s3-us-west-2.amazonaws.com\/courses-images\/wp-content\/uploads\/sites\/198\/2016\/11\/28190207\/Figure_13_03_04.jpg\" alt=\" Photo shows an orange daylily\" width=\"349\" height=\"262\" \/> Figure\u00a04. (credit: Steve Karg)[\/caption]\r\n\r\nAn individual with more than the correct number of chromosome sets (two for diploid species) is called <span style=\"text-decoration: underline\">polyploid<\/span>. Fertilization of an abnormal diploid egg with a normal haploid sperm would yield a triploid zygote. Polyploid animals are extremely rare, with only a few examples among the flatworms, crustaceans, amphibians, fish, and lizards. Polyploid animals are sterile because meiosis cannot proceed normally.\u00a0 Instead, they produce mostly aneuploid daughter cells that cannot yield viable zygotes. Polyploidy is very common in the plant kingdom, where olyploid plants tend to be larger and more robust than euploids of their species (Figure\u00a04).\r\n<h2>Sex Chromosomes<\/h2>\r\nWhat, if anything, can go wrong with the sex chromosomes?\u00a0 Rather than a gain or loss of autosomes, variations in the number of sex chromosomes are associated with relatively mild effects.\u00a0 This occurs because of a molecular process called X inactivation. Early in development when female mammalian embryos consist of just a few thousand cells, one X chromosome in each cell inactivates by tightly condensing into a dormant structure called a Barr body. The chance that an X chromosome, from either parent, is inactivated in each cell is random, but once the inactivation occurs, all cells derived from that one will have the same inactive X chromosome or Barr body. (credit: Michael Bodega)[\/caption]\r\n\r\nThis can be seen in so-called \u201ctortoiseshell\u201d cats where embryonic X inactivation is observed as color variegation (Figure\u00a05).\u00a0 Heterozygous females for an X-linked coat color gene will express one of two different coat colors over different regions of their body, corresponding to whichever X chromosome is inactivated in the embryonic cell\u00a0 of that region.\r\n\r\nAn individual with an abnormal number of X chromosomes will inactivate all but one X chromosome in each of her cells. However, even inactivated X chromosomes continue to express a few genes.\u00a0 As a result, X-chromosomal abnormalities are associated with mild mental and physical defects and sterility.\u00a0 Complete absence of the X chromosome prevents an individual from developing in utero.\r\n\r\nSeveral errors in sex chromosome number have been noted.\r\n\r\n(1)\u00a0 Triplo-X female - XXX female; express developmental delays; reduced fertility\r\n\r\n(2) \u00a0 Klinefelter syndrome - XXY male; small testes; enlarged breasts; reduced body hair\r\n\r\n(3)\u00a0 Turner syndrome -\u00a0 X0 female; (only a single sex chromosome); short stature; webbed skin in neck region; hearing and cardiac impairments; and sterility\r\n<h2>Duplications and Deletions<\/h2>\r\n[caption id=\"attachment_1472\" align=\"alignright\" width=\"350\"]<img class=\" wp-image-1472\" src=\"https:\/\/s3-us-west-2.amazonaws.com\/courses-images\/wp-content\/uploads\/sites\/198\/2016\/11\/28190330\/Figure_13_03_06.jpg\" alt=\"Photos show a boy with cri-du-chat syndrome. (credit: Paola Cerruti Mainardi)[\/caption]\r\n\r\nIn addition to the loss or gain of an entire chromosome, a chromosomal segment may be duplicated or deleted. Duplications and deletions produce offspring that survive but exhibit physical and mental abnormalities. Duplicated chromosomal segments may fuse to existing chromosomes or may be free in the nucleus. Cri-du-chat (from the French for \u201ccry of the cat\u201d) is a syndrome associated with nervous system abnormalities and identifiable physical features that result from a deletion of most of chromosome 5. (Figure\u00a06). Infants with this genotype emit a characteristic high-pitched cry from which the disordered is named.\r\n<h2>Structural Chromosome Rearrangements<\/h2>\r\nScientists have characterized numerous rearrangements in chromosome structure.\u00a0 The most common of which are chromosome inversions and translocations.\u00a0 Both are identified during meiosis.\u00a0 If the genes carried on homologous chromosomes are not oriented correctly, a recombination event could result in a loss of genes from one chromosome and the gain of genes on the other.\r\n<h3>Chromosome Inversions<\/h3>\r\nA chromosomal inversion is the detachment with a 180\u00b0 rotation and reinsertion of a chromosome part.\u00a0 Unless they disrupt a gene sequence, inversions only change gene orientation.\u00a0 Altered gene orientation can result in functional changes due to gene regulators being moved out of position with respect to their targets.\u00a0 Abnormal levels of gene products could result.\r\n\r\nAn chromosomal inversion can be pericentric, including the centromere, or paracentric, occurring outside of the centromere (Figure\u00a07). A pericentric inversion that is asymmetric can change the relative lengths of the chromosome arms, making these inversions easily identifiable.\r\n\r\n[caption id=\"attachment_1473\" align=\"aligncenter\" width=\"544\"]<img class=\"size-full wp-image-1473\" src=\"https:\/\/s3-us-west-2.amazonaws.com\/courses-images\/wp-content\/uploads\/sites\/198\/2016\/11\/28190419\/Figure_13_03_07.jpg\" alt=\" Illustration shows pericentric and paracentric inversions. A pericentric inversion can change the relative lengths of the chromosome arms; a paracentric inversion cannot.[\/caption]\r\n\r\nWhen one homologous chromosome undergoes an inversion but the other does not, the individual is described as an inversion heterozygote. To maintain point-for-point synapsis during meiosis, one homologous chromosome must form a loop, while the other molds around it. \u00a0 This topology can ensure that the genes are correctly aligned but forces stretching and can be associated with regions of imprecise synapsis (Figure\u00a08).\r\n\r\n[caption id=\"attachment_1474\" align=\"aligncenter\" width=\"544\"]<img class=\"size-full wp-image-1474\" src=\"https:\/\/s3-us-west-2.amazonaws.com\/courses-images\/wp-content\/uploads\/sites\/198\/2016\/11\/28190456\/Figure_13_03_08.jpg\" alt=\" This illustration shows the inversion pairing that occurs when one chromosome undergoes inversion but the other does not. This inversion pairing is essential to maintaining gene alignment during meiosis and to allow for recombination.[\/caption]\r\n\r\n<div class=\"textbox key-takeaways\">\r\n<h3>Evolution Connection<\/h3>\r\n<h4>The Chromosome 18 Inversion<\/h4>\r\nNot all structural rearrangements of chromosomes produce nonviable, impaired, or infertile individuals. In rare instances, such a change can result in the evolution of a new species.\u00a0 A pericentric inversion in chromosome 18 appears to have contributed to the evolution of humans. This inversion is not present in our closest genetic relatives, the chimpanzees. Humans and chimpanzees differ cytogenetically by pericentric inversions on several chromosomes and by the fusion of two separate chromosomes in chimpanzees that correspond to chromosome #2 in humans.\r\n\r\nThe pericentric chromosome 18 inversion is believed to have occurred in early humans following their divergence from a common ancestor with chimpanzees approximately five million years ago. Researchers characterizing this inversion have suggested that approximately 19,000 nucleotide bases were duplicated on 18p, and the duplicated region inverted and reinserted on chromosome 18 of an ancestral human.\r\n\r\nA comparison of human and chimpanzee genes in the region of this inversion indicates that two genes\u2014<em>ROCK1<\/em> and <em>USP14<\/em>\u2014that are adjacent on chimpanzee chromosome 17 (which corresponds to human chromosome 18) are more distantly positioned on human chromosome 18. This suggests that one of the inversion breakpoints occurred between these two genes. Interestingly, humans and chimpanzees express <em>USP14<\/em> at distinct levels in specific cell types, including cortical cells and fibroblasts. Perhaps the chromosome 18 inversion in an ancestral human repositioned specific genes and reset their expression levels in a useful way. Because both <em>ROCK1<\/em> and <em>USP14<\/em> encode cellular enzymes, a change in their expression could alter cellular function. It is not known how this inversion contributed to hominid evolution, but it appears to be a significant factor in the divergence of humans from other primates.[footnote]Violaine Goidts et al., \u201cSegmental duplication associated with the human-specific inversion of chromosome 18: a further example of the impact of segmental duplications on karyotype and genome evolution in primates,\u201d <em>Human Genetics<\/em>. 115 (2004):116\u2013122[\/footnote]\r\n\r\n<\/div>\r\n<h3>Translocations<\/h3>\r\n[caption id=\"attachment_1475\" align=\"alignright\" width=\"450\"]<img class=\" wp-image-1475\" src=\"https:\/\/s3-us-west-2.amazonaws.com\/courses-images\/wp-content\/uploads\/sites\/198\/2016\/11\/28190533\/Figure_13_03_09.jpg\" alt=\" Illustration shows a reciprocal translocation in which DNA is transferred from one chromosome to another. (credit: modification of work by National Human Genome Research\/USA)[\/caption]\r\n\r\nA translocation occurs when a chromosomal segment detaches and reattaches to a different, nonhomologous chromosome. Translocations can be minor or have devastating effects, depending on the altered gene position with respect to regulatory sequences. Specific translocations have been associated with several cancers and with schizophrenia. Reciprocal translocations result from the exchange of chromosome segments between two nonhomologous chromosomes where there is no gain or loss of genetic information (Figure\u00a09).\r\n<h2>Section Summary<\/h2>\r\nThe number, size, shape, and banding pattern of chromosomes make them easily identifiable in a karyotype and allows for assessment of many chromosomal abnormalities. Disorders in chromosome number, or aneuploidies, are typically lethal to the embryo, although a few trisomic genotypes are viable. Because of X inactivation, Changes in sex chromosomes typically have milder phenotypic effects. Aneuploidies also include instances in which segments of a chromosome are duplicated or deleted. Chromosome disorders can be divided into two categories: abnormalities in chromosome number and chromosomal structural rearrangements. Because even small segments of chromosomes span many genes, chromosomal disorders are characteristically dramatic and often fatal.<\/p>\n<h2>Identification of Chromosomes<\/h2>\n<p>How does one find out about their genetic make-up?\u00a0 What diseases, if any, do I have?\u00a0 What sort of chromosomes do I pass on to my progeny?\u00a0 One way we can find answers to these and other genetic questions is to have a karyotype. \u00a0 A karyotype is the number and appearance of chromosomes, including their length, banding pattern, and centromere position. In a human karyotype, autosomes(non-sex) are generally organized in approximate order of size from largest (chromosome 1) to smallest (chromosome 22). The X and Y chromosomes are not autosomes.\u00a0 During the naming of Down syndrome to trisomy 21, it was discovered that chromosome 21 is actually shorter than chromosome 22.\u00a0 Not wanting to change the name of this important disease, chromosome 21 retained its numbering, despite being the shortest set of chromosomes. The chromosome \u201carms\u201d projecting from either end of the centromere may be designated as short or long, depending on their relative lengths. The short arm is abbreviated <em>p <\/em>(for \u201cpetite\u201d), whereas the long arm is abbreviated <em>q <\/em>(because it follows \u201cp\u201d alphabetically). Each arm is further subdivided and denoted by a number. Using this naming system, locations on chromosomes can be described consistently in the scientific literature.<\/p>\n<div class=\"textbox key-takeaways\">\n<h3>Career Connection<\/h3>\n<h4>Geneticists Use Karyograms to Identify Chromosomal Aberrations<\/h4>\n<p>Although Mendel is referred to as the \u201cfather of modern genetics,\u201d he performed his experiments with none of the tools that the geneticists of today routinely employ. One such powerful cytological technique is karyotyping, a method in which traits characterized by chromosomal abnormalities can be identified from a single cell. To observe an individual\u2019s karyotype, a person\u2019s cells (like white blood cells) are first collected from a blood sample or other tissue. In the laboratory, the isolated cells are stimulated to begin actively dividing. A chemical\u00a0 is then applied to cells to arrest condensed chromosomes in metaphase. Cells are then made to swell using a hypotonic solution so the chromosomes spread apart. Finally, the sample is preserved in a fixative and applied to a slide.<\/p>\n<p>The geneticist then stains chromosomes with one of several dyes to better visualize the distinct and reproducible banding patterns of each chromosome pair. Following staining, the chromosomes are viewed using bright-field microscopy. A common stain choice is the Giemsa stain. Giemsa staining results in approximately 400\u2013800 bands (of tightly coiled DNA and condensed proteins) arranged along all of the 23 chromosome pairs.\u00a0 An experienced geneticist can identify each band. In addition to the banding patterns, chromosomes are further identified on the basis of size and centromere location. To obtain the classic depiction of the karyotype in which homologous pairs of chromosomes are aligned in numerical order from longest to shortest, the geneticist obtains a digital image, identifies each chromosome, and manually arranges the chromosomes into this pattern (Figure 1).<\/p>\n<p>At its most basic, the karyogram may reveal genetic abnormalities in which an individual has too many or too few chromosomes per cell. Examples of this are Down Syndrome, which is identified by a third copy of chromosome 21, and Turner Syndrome, which is characterized by the presence of only one X chromosome in women instead of the normal two. Geneticists can also identify large deletions or insertions of DNA. For instance, Jacobsen Syndrome, involving distinctive facial features as well as heart and bleeding defects, is identified by a deletion on chromosome 11.\u00a0 The karyotype can pinpoint translocations, which occur when a segment of genetic material breaks from one chromosome and reattaches to another chromosome or to a different part of the same chromosome. Translocations are implicated in certain cancers, including chronic myelogenous leukemia.<\/p>\n<p>During Mendel\u2019s lifetime, inheritance was an abstract concept that could only be inferred by performing crosses and observing the traits expressed by offspring. \u00a0 Today\u2019s geneticists can actually visualize the chromosomal composition of an individual to confirm or predict genetic abnormalities in offspring, even before birth.<\/p>\n<\/div>\n<h2>Disorders in Chromosome Number<\/h2>\n<p>Abnormalities in chromosome number are the most obviously identifiable form of a karotype.\u00a0 Most of these are due to nondisjunction.\u00a0 <span style=\"text-decoration: underline\">Nondisjunction<\/span> occurs when homologous chromosomes pairs or sister chromatids fail to separate during meiosis. Causes that attribute to nondisjunction are misaligned or incomplete synapsis or a dysfunction of the spindle apparatus, facilitating chromosome migration. Nondisjunction rates increase with parental age.<\/p>\n<p>Nondisjunction can occur during either meiosis I or II, with differing results (Figure\u00a02). If homologous chromosomes fail to separate during meiosis I, the result is two gametes lacking a that particular chromosome and two gametes with two copies of the chromosome. An individual with an error in chromosome number is described as<span style=\"text-decoration: underline\"> aneuploid<\/span>, a term that includes monosomy (loss of one chromosome) or trisomy (gain of an extraneous chromosome). Monosomy zygotes, missing any one copy of an autosome, fail to develop to birth because they lack essential genes.\u00a0 Most autosomal trisomies fail to develop to birth; however, duplications of some of the smaller chromosomes (13, 15, 18, 21, or 22) can result in offspring that can survive.\u00a0 Trisomic individuals suffer from\u00a0 an excess in gene dose. Individuals with an extra chromosome may synthesize an abundance of the gene products encoded by that chromosome. This extra dose of specific genes can lead to a number of functional challenges, often precludes development. The most common trisomy among viable births is that of chromosome 21, or Down Syndrome. Individuals with this inherited disorder are characterized by short stature, stunted digits, facial distinctions including a broad skull and large tongue, and significant developmental delays. The incidence of Down syndrome is correlated with the age of the mother.\u00a0 Older women are more likely to become pregnant with fetuses carrying the trisomy 21 genotype (Figure\u00a03).<\/p>\n<h2>Polyploidy<\/h2>\n<div id=\"attachment_1470\" style=\"width: 359px\" class=\"wp-caption alignright\"><img loading=\"lazy\" decoding=\"async\" aria-describedby=\"caption-attachment-1470\" class=\"wp-image-1470\" src=\"https:\/\/s3-us-west-2.amazonaws.com\/courses-images\/wp-content\/uploads\/sites\/198\/2016\/11\/28190207\/Figure_13_03_04.jpg\" alt=\"Photo shows an orange daylily\" width=\"349\" height=\"262\" \/><\/p>\n<p id=\"caption-attachment-1470\" class=\"wp-caption-text\">Figure\u00a04. (credit: Steve Karg)<\/p>\n<\/div>\n<p>An individual with more than the correct number of chromosome sets (two for diploid species) is called <span style=\"text-decoration: underline\">polyploid<\/span>. Fertilization of an abnormal diploid egg with a normal haploid sperm would yield a triploid zygote. Polyploid animals are extremely rare, with only a few examples among the flatworms, crustaceans, amphibians, fish, and lizards. Polyploid animals are sterile because meiosis cannot proceed normally.\u00a0 Instead, they produce mostly aneuploid daughter cells that cannot yield viable zygotes. Polyploidy is very common in the plant kingdom, where olyploid plants tend to be larger and more robust than euploids of their species (Figure\u00a04).<\/p>\n<h2>Sex Chromosomes<\/h2>\n<p>What, if anything, can go wrong with the sex chromosomes?\u00a0 Rather than a gain or loss of autosomes, variations in the number of sex chromosomes are associated with relatively mild effects.\u00a0 This occurs because of a molecular process called X inactivation. Early in development when female mammalian embryos consist of just a few thousand cells, one X chromosome in each cell inactivates by tightly condensing into a dormant structure called a Barr body. The chance that an X chromosome, from either parent, is inactivated in each cell is random, but once the inactivation occurs, all cells derived from that one will have the same inactive X chromosome or Barr body. (credit: Michael Bodega)<\/p>\n<\/div>\n<p>This can be seen in so-called \u201ctortoiseshell\u201d cats where embryonic X inactivation is observed as color variegation (Figure\u00a05).\u00a0 Heterozygous females for an X-linked coat color gene will express one of two different coat colors over different regions of their body, corresponding to whichever X chromosome is inactivated in the embryonic cell\u00a0 of that region.<\/p>\n<p>An individual with an abnormal number of X chromosomes will inactivate all but one X chromosome in each of her cells. Duplications and deletions produce offspring that survive but exhibit physical and mental abnormalities. Duplicated chromosomal segments may fuse to existing chromosomes or may be free in the nucleus. Cri-du-chat (from the French for \u201ccry of the cat\u201d) is a syndrome associated with nervous system abnormalities and identifiable physical features that result from a deletion of most of chromosome 5. (Figure\u00a06). Infants with this genotype emit a characteristic high-pitched cry from which the disordered is named.<\/p>\n<h2>Structural Chromosome Rearrangements<\/h2>\n<p>Scientists have characterized numerous rearrangements in chromosome structure.\u00a0 The most common of which are chromosome inversions and translocations.\u00a0 Both are identified during meiosis.\u00a0 If the genes carried on homologous chromosomes are not oriented correctly, a recombination event could result in a loss of genes from one chromosome and the gain of genes on the other.<\/p>\n<h3>Chromosome Inversions<\/h3>\n<p>A chromosomal inversion is the detachment with a 180\u00b0 rotation and reinsertion of a chromosome part.\u00a0 Unless they disrupt a gene sequence, inversions only change gene orientation.\u00a0 Altered gene orientation can result in functional changes due to gene regulators being moved out of position with respect to their targets.\u00a0 Abnormal levels of gene products could result.<\/p>\n<p>An chromosomal inversion can be pericentric, including the centromere, or paracentric, occurring outside of the centromere (Figure\u00a07). A pericentric inversion that is asymmetric can change the relative lengths of the chromosome arms, making these inversions easily identifiable.<\/p>\n<div id=\"attachment_1473\" style=\"width: 554px\" class=\"wp-caption aligncenter\"><img loading=\"lazy\" decoding=\"async\" aria-describedby=\"caption-attachment-1473\" class=\"size-full wp-image-1473\" src=\"https:\/\/s3-us-west-2.amazonaws.com\/courses-images\/wp-content\/uploads\/sites\/198\/2016\/11\/28190419\/Figure_13_03_07.jpg\" alt=\"Illustration shows pericentric and paracentric inversions. A pericentric inversion can change the relative lengths of the chromosome arms; a paracentric inversion cannot.<\/p>\n<\/div>\n<p>When one homologous chromosome undergoes an inversion but the other does not, the individual is described as an inversion heterozygote. To maintain point-for-point synapsis during meiosis, one homologous chromosome must form a loop, while the other molds around it. \u00a0 This topology can ensure that the genes are correctly aligned but forces stretching and can be associated with regions of imprecise synapsis (Figure\u00a08).<\/p>\n<div id=\"attachment_1474\" style=\"width: 554px\" class=\"wp-caption aligncenter\"><img loading=\"lazy\" decoding=\"async\" aria-describedby=\"caption-attachment-1474\" class=\"size-full wp-image-1474\" src=\"https:\/\/s3-us-west-2.amazonaws.com\/courses-images\/wp-content\/uploads\/sites\/198\/2016\/11\/28190456\/Figure_13_03_08.jpg\" alt=\"This illustration shows the inversion pairing that occurs when one chromosome undergoes inversion but the other does not. This inversion pairing is essential to maintaining gene alignment during meiosis and to allow for recombination.<\/p>\n<\/div>\n<div class=\"textbox key-takeaways\">\n<h3>Evolution Connection<\/h3>\n<h4>The Chromosome 18 Inversion<\/h4>\n<p>Not all structural rearrangements of chromosomes produce nonviable, impaired, or infertile individuals. In rare instances, such a change can result in the evolution of a new species.\u00a0 A pericentric inversion in chromosome 18 appears to have contributed to the evolution of humans. This inversion is not present in our closest genetic relatives, the chimpanzees. Humans and chimpanzees differ cytogenetically by pericentric inversions on several chromosomes and by the fusion of two separate chromosomes in chimpanzees that correspond to chromosome #2 in humans.<\/p>\n<p>The pericentric chromosome 18 inversion is believed to have occurred in early humans following their divergence from a common ancestor with chimpanzees approximately five million years ago. Researchers characterizing this inversion have suggested that approximately 19,000 nucleotide bases were duplicated on 18p, and the duplicated region inverted and reinserted on chromosome 18 of an ancestral human.<\/p>\n<p>A comparison of human and chimpanzee genes in the region of this inversion indicates that two genes\u2014<em>ROCK1<\/em> and <em>USP14<\/em>\u2014that are adjacent on chimpanzee chromosome 17 (which corresponds to human chromosome 18) are more distantly positioned on human chromosome 18. This suggests that one of the inversion breakpoints occurred between these two genes. Interestingly, humans and chimpanzees express <em>USP14<\/em> at distinct levels in specific cell types, including cortical cells and fibroblasts. Perhaps the chromosome 18 inversion in an ancestral human repositioned specific genes and reset their expression levels in a useful way. Because both <em>ROCK1<\/em> and <em>USP14<\/em> encode cellular enzymes, a change in their expression could alter cellular function. Translocations can be minor or have devastating effects, depending on the altered gene position with respect to regulatory sequences. Specific translocations have been associated with several cancers and with schizophrenia. Reciprocal translocations result from the exchange of chromosome segments between two nonhomologous chromosomes where there is no gain or loss of genetic information (Figure\u00a09).<\/p>\n<h2>Section Summary<\/h2>\n<p>The number, size, shape, and banding pattern of chromosomes make them easily identifiable in a karyotype and allows for assessment of many chromosomal abnormalities. Disorders in chromosome number, or aneuploidies, are typically lethal to the embryo, although a few trisomic genotypes are viable. Because of X inactivation, Changes in sex chromosomes typically have milder phenotypic effects. Aneuploidies also include instances in which segments of a chromosome are duplicated or deleted.
